Visit Gaudis
Eat Tapas
Do walking tours (barrio Gotic, Picasso, etc)
Take a day trip - we did Girona which is worthwhile, but there are many other possibilities as well

I wouldn't go to Barcelona and do just one thing. But in addition to those listed above, I would go to a performance at the Palau de la Musica Catalana. And I would go to the museum of the history of the city of Barcelona and walk through the underground remains of the Roman city.
And I would eat as many kinds of odd looking clams as I could find.

Nikki, I remember your report about the clams. I don't know if I have ever eaten clams. I am not going to do just one thing. I am going to do all the one things that people tell me about. So far, everything I see is already on my list. Check my other post.

Narrowing it down to 1 thing is just impossible. Tops:
Palau de la Musica Catalana (tour and/or concert)
Sagrada Familia
Santa Maria del Mar (this might not be on many people's tops lists, but I just loved this church)
quick train ride to Colonia Guell to see the crypt
Sitting in a cafe or bar soaking up the atmosphere
Wandering, wandering, wandering the different great neighborhoods
And so much more...

Sagrada Familia.
I know it's a popular choice, but it was an easy one for me. As denon said, even if you don't love it, it's unique. There isn't anything else like it (not that I've ever seen anyway).
The other Gaudi sites would be next: Parc Guell, Casa Batllo, etc. And La Boqueria is interesting too.
