Presumably the layover is at the airport. Take the shuttle (S-Bahn) train to the Frankfurt main Station (Hauptbahnhof); walk or take a tram or subway to the Hauptwache station. From there walk toward the Main river and visit the old (actually rebuilt) part of the city - - - Roemer City Hall, Cathedral, etc.
Then cross the river on the pedestrian bridge (Eiserner Steg)and walk downriver on the opposite (Sachsenhausen) side of the river. There are several good museums along the river promenade. Then return to the main station by crossing the lower bridge.

Frankfurt is one of the ugliest cities in Germany. Trains from the airport will take you elsewhere - along the Rhine, towards Heidelberg - one of the nicest German cities. the above walk in above post is a great one if bent on Frankfurt as it takes you by the city's few neat old (pieced together from WWII rubble) things. Not really that Frankfurt can't be enjoyable - has great museums for one thing, just that there are much better places by direct train from airport. Frankfurt is europe's banking capital and it has taken on, unusual for Europe, the look of a prosperous American city.
