Yeah, I thought the OP meant the 40L pack as a day bag too - for sightseeing and so on. That seems really big to me. No need to carry that large of a bag around.
I would pack a slightly larger suitcase or duffel (still within the carryon limits) and then a smaller day bag - small backpack or messenger bag). That will be much easier to handle on planes and trains and also leaves you with a more versatile day bag.

Might also ask if the rolling duffel being proposed has a long enough handle for the OP to pull without "stooping"--that would get really uncomfortable with a backpack on. Test the entire travel kit.
