I would suggest (as has been mentioned on another thread here recently) using the Queenston-Lewiston bridge to cross into Canada. I'm not familiar with the location of the airport, so other people might have other ideas, but the Queenston-Lewiston bridge is generally less busy than the Peace Bridge or the Rainbow Bridge. You want to get on 190 north to access it. Once you've crossed, the Queen Elizabeth Highway is right there and will take you right into downtown Toronto. You'd be best advised to have a passport with you; if you don't have one, they'll accept proof of citizenship (like a birth certificate) and photo ID. I think they want 2 pieces of that. It's a ten minute drive from the airport to the Peace Bridge in downtown Buffalo. From the airport, turn right onto Genesee Street (or left, if you're coming from one of the rental car sites across from the airport), and turn onto the Kensington Expressway (Rt. 33). Take this right into downtown, and it connects to the 190. Take the 190 west for about 1/2 a mile to the Peace Bridge exit. Once you cross the bridge, it straight up the QEW to Toronto - a 90 minute drive. Sometimes the Peace Bridge is busy, sometimes you drive right over in 5 minutes. You must have an original birth certificate and a photo I.D. (driver's license). Have a great trip.

As you are coming from Buffalo, there are several attractions along the way that some of our friends will stop at before heading into Toronto. Some do Niagara Falls on the way in or on the way back, or the Lewiston Outlet Malls, or some of the wonderful vineyards like Vineland Estates. Just a thought.
