Cheapest is either drive or Greyhound. It's about 1,300 miles, so you need two nights of hotels - which may add to the cost. If you take the bus, it's still almost 48hours, and I know all of you will scream.
The quickest way is to fly. Air Canada has connection in Toronto.

Air Canada flies to Columbus (travelled to there to work at McGraw Hill for a few weeks) and to Charlottetown PEI.
Another option is to fly to to Boston by the cheapest way possible and take Air Canada from Boston to Halifax ($159) and start your journey from there or take another flight to Charlotteown from Halifax. It's about a two-three hour drive to PEI.
There are also flights from Montreal and Ottawa to Charlotteown.
