1-1/2 hours max. to Niagara Falls.

if you want to avoid toll roads....
I'm not sure of the exact road name coming right out of the airport (DIXIE ROAD - south/east, I believe).
Immediately get on
HWY 401 WEST.
then immediately look for & get on
HWY 403 towards Hamilton.
Continue past Hamilton, over the Burlington Skyway (just a long bridge).
Signs to HIGHWAY - Q.E.W.
Road now becomes the
Q.E.W. (Queen Elizabeth's Way) highway.
Continue to Niagara Falls! Lots of signs. Easy to find.
HWY #420 once you get really close to Niagara Falls.

The fact that you asked for a timetable gives me the impression you don't want to drive.
There is a shuttle bus service that connects Pearson and Niagara, which are 78 miles (125 kilometers) from each other:

if you are driving, from the airport (it's about 80 miles or 120 KM) take the 427 south to the QEW towards (West). Then you will stay on the QEW for atleast 90 minutes depending on how fast you drive. Just make sure that you always stay on the QEW becasue just after Oakville there is a fork in the road, stay left because if you go right then you are heading to Hamilton on the 403. Then your screwed ;-). It's really an easy drive!
