My daughter wants to honeymoon for a few days in Amalfi. What is the best way to get there from Florence or from Rome. I assume they will have to use Naples. They will want to experience the drive at least one way and stay there for 2 days. Can they ferry back to Naples or Rome from there?

Do you mean the city of Amalfi, or just the area?
What time of year?
Your question is more complex than you may know.

If you are talking about the town of Amalfi, the easiest way is train to Salerno and then ferry from Salerno to Amalfi. On the way back they can take the bus or a driver from Amalfi to Salerno and then train to Rome or Naples.

I second ekc's suggestion.

The ferry dock is only a couple of blocks from the train station. The ride is very scenic.

Amalfi is quite interesting.

They don't have to take a limo from Naples to the AC.
The Circumvesuviana commuter train will bring them to Sorrento. The SITA bus will take them to Positano, etc.

What drive? From Rome to Sorrento? The AC coast road?

What is the rest of this itinerary? Two days on the AC is rather rushed.

If they are going to Florence, why not visit the Cinque Terre instead of the AC?

Hi Ira, talking about Vesuviana and sita, have you experienced it on your own????? Of course, it is less expensive, but depends what they are looking for.
well, on a honeymoon trip, you probably want to relax. Think about this: once arrived to the train station of Naples, wait for the Circumvesuviana connection (up to 20 minutes), 1,20 min of ride till Sorrento (the train can be full, so the seats are not always available), once arrived to Sorrento, take a bus to Positano (another 30 minutes, or to Amalfi (another hour). don't forget about the luggage you have to transport! Moreover, to get to the amalfi coast, you don't have to get untill Sorrento (have a look at the map), because the turn to the amalfi coast is at the Meta di Sorrento (much before the Sorrento) . so, basicaly, once arrived to Sorrento, they will have to take a bus that will drive them back to a certain point to, then, take a road to the amalfi coast. again, remember that on the pubblic transportation the seats are not guaranteed and the busses ara packed in hight season.
So, why to waste preciuos time and energy on the vacation?

If going to Amalfi using public trans, the route through Salerno is more pleasant than the route through Sorrento, especially in summer/high season.

"They will want to experience the drive at least one way..." Which drive? The Amalfi Coast, Florence-Amalfi, Rome-Amalfi? It doesn't sound like they have a whole lotta time for long drives.

If they want to see Pompeii during their 2 days, careful planning will be required. Where are they going after Amalfi?

Yes, I have done the Circumvesuviana and the SITA bus from Naples to Sorrento.

I did not tell them that that was what they should do, only that it was an alternative.

Maybe 110E each way is a bit high for them.

I am not sure you meant Amalfi or Amalfi area. In second case I would rather focus on a nice resort with an attentive hospitality being honeymoon such a special stay. Distances are short so after electing the base you can visit all Amalfi Coast cities. Bob asked about the time of year because the Amalfi coast in a summer destination. Hotels and restaurants and ferries close down in the fall, reopen in later spring.

The Amalfi coast is a bit difficult to get to and get back from. They'll spend so much time in transportation that 2 nights/1 day is hardly worth it.

Ira's suggestion of the Cinque Terre is a good one, the five towns being closer to Florence and a little easier to get to. And fewer days in the CT is appropriate. There's not much to do but walk between the towns (or take the train or the ferry). But the CT is also a good weather destination. Look up Cinque Terre in Google Images.
