You may want to check out the guidebook "Off the Beaten Path in Pennsylvania". We used it when we went to Hershey/Lancaster. Had lots of listings for things we would never have found otherwise.
A friend took a similar trip but from Boston and heading west. Stopped at Baseball Hall of Fame in Cooperstown, Crayola factory, Hershey, saw a baseball game in Pittsburg. Went to Kings Island amusement park too. They loved the trip. Their kids were 9 and 7 at the time.

Depends on the time of year.
If summer, be sure to spend a day at Cedar Point, (Sandusky, Ohio) the amusement park with the most and best roller coasters in the WORLD (RE: Roller Coaster Magazine)
A visit to the Rock & Roll Hall of Fame (Cleveland) is fun at any time of year.

Cleveland's University Circle is a gem. You'll find world-class museums there -- the Cleveland Museum of Art, musueum of natural history (read: dinosaurs), botanical gardens, institutes of music and art, the Crawford Auto-Aviation Museum, and the campus of Case Western Reserve University. The Children's Museum is being renovated. Rockefeller Park is adjacent to the museums. UC sits next to the city's Little Italy district, which, as would expect, features terrific Italian cafes, shops, and art galleries.

Depending on the age of your kids, Conneaut Lake Park in NW PA might work. It's a real old-fashioned amusement park in a small town. If your kids are older, then they probably wouldn't enjoy it as much, but younger kids seem to love it. As a bonus, it's inexpensive.
Also, for a day at the beach, Presque Isle in Lake Erie is always a good option. And you're close to Waldameer Park--it's bigger than Conneaut Lake Park, but still not on the scale of Cedar Point.

Dayton, Ohio has an air and space museum, and a historical village with, among other things, Wright Brothers stuff.
In the summer, Kings Island amusement/water park, near Cincinnati, is fun.
My kids loved Gettysburg.
