Yes, a (long) day is probably enough for Baltimore's Inner Harbor. Be sure to visit the National Aquarium, and take in the view from Federal Hill on the south side of the Harbor. There's also a small Civil War Museum at the President Street station nearby. And the B&O Railroad Museum is a short cab ride away.
If you've got a car, it certainly isn't a bad drive. The Inner Harbor is readily accessible from I-95.
There are two ways to get there by train:
(1) Take a MARC (Maryland Commuter) train or Amtrak from Union Station, DC to Penn Station, Baltimore. You'll then have to take a cab from Penn Station to the Inner Harbor -- it's maybe 15 blocks.
(2) Take a MARC train from Union Station to Camden Station, Baltimore. This gets out in the shadow of the Camden Yards baseball stadium. You can then walk from there to the Inner Harbor (5 or 6 blocks).
Of course, the commuter trains run a lot less on weekends, so these are most attractive as an alternative during the week.

Driving will be the quickest way; if the traffic gods are with you, the trip will take less than an hour. It's probably the best way to get there from Alexandria.
A day is definitely enough to see the Inner Harbor. Make reservations to go to the Aquarium, it's great.
And if you have the time, visit Fells Point or go to Little Italy for a meal. Both are 5 minutes from the harbor

The National Aquarium is in Washington DC and is not worth the time. The Baltimore Aquarium is in the inner harbor, and is definitely worth the trip! If you drive up (about 45 minute drive), try to leave after 9-9:30am and don't return between 4-6:30pm to avoid rush hour. A 45 minute car ride will turn into a 1.5 hour one. Car is the most direct, fastest way to go, but the train is a great option. The MARC train runs on weekdays, and the Camden Line goes from Union Station to Camden Yards (2 block walk from inner harbor). If you take the Penn Line, I recommend that you take it from Union Station to BWI, then switch to light rail into Camden Yards, and walk from there. You'll have a great time. Go to www.mtamaryland.com for train/light rail/subway & bus info.
