Jljb - Your best source of info is the Rome tourist office website. It has an itinerary for seeing the best of Rome in 2 days:

We found this website and thought it was helpful in planning our 2 day visit. They have 48 hour and 96 our suggested tour. Hope it helps you. Enjoy your trip!

I think before you go on you should review your interests and hotel location.
That will have a lot to do with how you put your days together.
Whether or not you are a museum goer or a site walker makes a big difference.
We were in Rome three years ago and walked a good part of the city each day.
We also limited our museums to the Vatican Museum.
