Tuscany is a very large area. Better to state your budget and where you want to be..

Only one night? Where are you coming from and where are you going after that one night? Will you have a car, or are you traveling by train?

I might be inclined to pick a place that's less convenient to see by public trans, like Volterra, the Montalcino-SanQuirico-Pienza area or somewhere in Chianti. If you're spending several days in Florence, you can easily day-trip to Siena and/or San Gimignano. Volterra or Chianti would be an all-day drive if you stick to secondary roads (which I prefer), but you'd drive part of the shore of Lake Bolsena and pass through San Quirico. When is your trip?
