Have you been to Constanta? There are plenty to see there. Also, how much time do you have in port? Why would you spend 6 hours round trip in a car/bus/van/train/whatever to go to Bucharest?
The drive is more like 3 hours than 2.5 each way. if you hit traffic, it will be even longer.
IMHO, I wouldn't do it.

We took a tour of the Danube River Delta when visiting Constanta several years ago.It is a UNESCO world heritage site, and was fascinating to us. Thank goodness for digital media!
If you are not into nature tours, photography, or interested in the local culture, avoid this. About a third of our group of 12 found it boring.

We have never been to Constanta and in reading several people were disappointed in the port that did it on their own. Some of said they bit the bullet and did the drive to Bucharest. I have also heard about many people getting mugged in Constanta (maybe locals waiting for the ships to come into port). We are hiring a guide. She said it is a walking tour...
