From Memphis I would take the Natchez Trace down & skip Birmingham. In lieu of Jackson go to Natchez, MS. A great little town on the river w/loads of mansions. Not sure where you are starting from but if you would like more in depth info let me know. You can pick up the Trace in Tupelo (stop here if you like Elvis, it's his birthplace).

Memphibian here...If you are starting in Memphis, you have an option of going to New Orleans via I55 through Jackson, Ms (half way to New Orleans). N.O. is 400 miles from Memphis. If not stopping in Jackson, it is a sox hour drive.
If going to Birmingham, it is toward the southeast, which is great if you are going to Atlanta, but out of the way for Jackson, MS or New Orleans.

Drive down the River Road (River Highway?) and stop in Greenville, MS. There is not too much there, but you MUST eat at Doe's Eat Place and listen to some blues. Jackson - Cock of the Walk on the reservoir was fun, as was the museum (can't remember the name) downtown where we saw a great Eudura Welty exhibit. Don't know if the exhibit was permanent. Sorry for my lack of detail - I need to find my trip journal before posting I guess. Have fun! That area is great.

of all the places you cited, New Orleans is the only must do. As a native Mississippian, I think Natchez is beautiful to drive through but generally overrated. Vicksburg has more historical interest as far as river towns go.
Do you plan to stop along the way? A night in an antebellum B&B is probably your best bet.
Plan for more time in New Orleans than the rest of your stops. There's more to see and do there. Probably better food per square mile in the French Quarter than anywhere else in US, with possible exception of NYC.
Oxford, MS, is a nice little town about 30 minutes off the interstate south of Memphis. Great bookstore there, on the town square with other boutiques and such.
Jackson, MS, is home to one of the best bookstores in the world. Lemuria Bookstore. If you enjoy books and the reading world, it's definitely worth a stop.
I'm not well-versed on Memphis. But the barbecue there is overrated. You need to look for little hole in the walls in the Mississippi Delta for truly good bbq.
Lots of small towns in MS and LA have various museums, juke joints, antebellum home tours and such that are off the beaten path. If you're interested in anything along those lines, email me for further info.
When in New Orleans, stay in the French Quarter. Prices are lower there during the summer. The food is great, the weather almost unbearably hot, but the atmosphere makes it worth the sweat. You'll learn how to slow down and enjoy simple things like fine jazz, fine bourbon, and the blessings of ceiling fans, air conditioning, and rocking chairs.

I suppose the canals are over-rated in Venice too. I've found that Jackson, MS is kind of like Dayton, Ohio, only without the excitement but I'm not well versed either. Neely's on Jefferson and Interstate are my personal favorites but there are dozens of great BBQ joints to choose from in Memphis. Ask a local and don't eat bbq on Beale St. and you'll do fine.
