Well, you can't drive there and back in a day, unless you are a masochist. It is 450 miles, one way, from NYC to Niagara Falls. There are tour operators that will fly you to Buffalo, take you on a tour of the Falls, then fly you back. Cost is $300 - $350 per person. Here's one: http://www.bedoretours.com/international.htm#NYCTour

I just checked JetBlue Airlines from JFK to Buffalo and chose a random date of October 9th (a Friday) and you can fly for $44.00 each way + taxes. You can be in Buffalo by 8:00 a.m. You'd have to factor in taxi fares to and from both airports.

That's a great deal, BuffaloGirl. And if they're only going for a day, or overnight trip, then returning to the same NY hotel, maybe they could carry an overnight bag, leaving the rest of their stuff at their hotel, and use the subway ($1.50/$2?) in NYC.
Judging from you name, "BG", can you offer any advice for getting to the Falls from Buffalo? Thanks.

Public transportation in Buffalo/Niagara is terrible. the airport is in Cheektowaga and a cab to Niagara Falls will probably be $20-$30 each way-still far cheaper than the tour operator deal. However, you might be able to rent a car just for the day for less than 2 cab fares, and then have the car at your disposal. I suspect there are tour operators locally that might go from the airport to the falls, but being local, it's not something I ever looked into. You might try some local convention and visitors websites for more into. One web site that might direct you to it is OfficialTravelGuide.com. October is a great time to see the Falls-the leaves will be changing colors and its a little early for snow here.

Check out http://www.buffaloairporttaxi.com/
It's a $25 cab ride each direction. They also have tours.
Rachel is correct...public transportation is lousy here. However, if you only have a day and don't want to deal with a car, then a taxi is the best way. Wear a good pair of walking shoes and you can walk to all the vantage points on the American side. You can even walk over the bridge to the Canadian side. Enjoy.

I forgot to mention that there is a shuttle that I "think" is free on the American side and does the drop off/pick up thing along the various viewing sites so you won't have to walk it all. Your taxi driver will know where to drop you off to pick up the shuttle.
