My girlfriends and I took a chick trip to Charleston, South Carolina, and it was quite nice. We toured the old plantations and went on a candlelight tour of homes - really fun. Also great seafood and beautiful gardens in the area.
San Francisco is really nice - loads to do, and you can take in the wineries if that interests you.

I second the suggestion of Charleston. The time of year is perfect. If your mother is a gardner, suggest you give her a copy of "Mrs Whaley's Charleston Gardens" as a gift to help her get in teh spirit of Charleston. It can be ordered from the Charleston Historical Society. A good base for you would be the Doubletree Suites. Have fun and spoil your mom!

Where are you leaving from? Even in mid America there are surely tons of places to go for a nice long-weekend. How far do you want to travel?
In any case, find a nice resort with a spa/salon so your mom can spend an afternoon being pampered. I'm not into massages, but I love a facial, mani and pedi. Then a dinner out at someplace really special.
