What is a POI?
If it is "point of interest", there are famous art museums in St Louis and Kansas City and two very nice smaller ones in Columbus, Ohio, the Columbus Museum of Art and the Wexner at the University, which is architecturally interesting, too. You could see the Arch in St Louis, but it is pretty meh.
Eat barbecue in Kansas City, ribs in St Louis. Ohio is the fattest state in the country, so you may just want to slide right through
but if you stop in Columbus, German Village is literally right on the edge of the interstate.
There are lots of corn and soybeans on the way. This trip, like any long distance trip, is vastly improved by driving part of the time on roads that go through small towns. Better food, funky local bars -- and museums. Stop in the public library to use the internet or ask what is interesting locally. US 50 is, I think, one of the major parallel highways on this route.

Just south of Wheeling, WV (on I-70) is the town of Moundsville and the former West Virginia Penitentiary which offers day tours, ghost tours and overnight tours.

Crossing Kansas on I-70, some of the diverse cool stops are the Grass Roots Arts Center & Garden of Eden in Lucas, Cathedral of the Plains in Victoria, and the Castle ROck Badlands in the open range land. Again, depending on your interests, there are a number of good museums.

The Arch is "pretty meh"?
My heart leaps up when I behold the Arch in the sky:
So was it when my life began;
So would Wordsworth feel if he were cruising down I64/US40 in his way from DC to Denver. If it's 104 in the shade, even he would at most make a short detour to see what it looks like from the street nearby.
