A lot of what I could suggest for Colorado involves the type of vehicle you will be driving. If you have an SUV type, more avenues are open.
For example, the most attractive area I know of in Colorado is the Horn Fork Basin of Cottonwood Creek, which is in the heart of the Saguache Range west of Buena Vista. Getting there involves driving over a very rough road. I made it once in a '71 Chevvy, but I had to get out and lie on my stomach to direct my wife over the rocks so that the transmission would survive. The Basin itself is nestled in a glacial valley with Mt. Harvard to the northwest, Mt. Columbia to the east, and Mt. Yale to the south, and the rocky pinnacles of the Columbia Harvard wall blocking your path to the north and northeast.
Other than that, there is Ouray and its hots springs. Near Ouray is Yankee Boy Basin that is beautiful. It is a high mountain valley surrounded by high peaks, including Mt. Sneffels.
The drive from Ouray to Durango over the Million Dollar highway (1 million per mile to construct years ago.)is stunning. Also, the Black Canyon of the Gunnison, Mesa Verde National Parks,
Rocky Mountain National Park, Colorado National Monument, and Pike's Peak are all prime attractions.
Tell me where you are flying into and out of and I can give some more suggestions. My son is out there now vacationing in Estes Park and I am sure he will have a few ideas, too.

I love the entire state of Colorado, but if I had to pick one spot, it would be Mesa Verde! So, with that in mind, my preference is the western part of the state. My dream trip would have me veturing into northeast Arizona as well. My list would be headed by Mesa Verde, Canyon de Chelly (in northeast Arizona) and Monument Valley (on the Arizona-Utah border). You could also include Chaco Canyon in northwest New Mexico, plus other sites in western Colorado.
I believe the closest airport in that area is Durango. I know that United flies there and you can connect from Denver.
