Bosque del Cabo has more hiking trails and presumably wildlife. There's a semi-decent chance of seeing a puma there.
If you want the option of diving, Drake provides access to Cano Island, which is probably the best diving on/near the mainland (Cocos is a different category of course).
Can't go wrong.

Bosque del Cabo has the most wildlife that I've seen with seven trips to Costa Rica in many different areas. But, if diving is still one of your main interests, then I would opt for Drake Bay and a trip to Cano Island.

Have not stayed there, but have read good reviews. Supposed to be a lot of wildlife on the property - howlers, scarlet macaws, etc. From what I've read, it's about a 15 minute walk to get to restaurants, so most people do their own cooking on the property. Supposed to be rustic but comfortable. If you go, I'd love to hear what you thought.

We walked along the road by Casa Bambu and saw the elusive squirrel monkeys along with spider monkeys. I haven't stayed there, but have also read good reviews of it. I think I would want a car as it's a bit out of town. From what I remember it's within walking distance to Martina's, but there's not much else as far as restaurants, etc.
