Venice Transport
I am planning a trip to multiple cities in Italy and will be going to Venice (VCE MARCO POLO) first. Since it is the first time there, I would like to know the best way to get from the airport to city. My package came with tickets from Venezia St Lucia to Venezia Mestre.

There is regular vaporetto (public boat) service from Pzle Roma to many parts of Venice.
Investigate buying a boat pass if in the city long enough - even a day pass - be sure to float down the Grande Canale at night - to me the most sublime experience in Europe.

We have done the water taxi and just last week did the bus and vaporetta way. the bus/vaporetta could not have been easier!
The bus is the blue bus and drops off and picks up right outside the terminal! With the water taxi, you have to walk around the terminal and down to the docks - not far but a little disconcerting when you don't know where you're going. Also, we found cuing up for the taxi took over 1/2 hr.
The bus was 3E each. At Piazzale Roma, walk from the bus stop towards the canal and the vaporetta stop is right there. As PalenQ said, depending on how long you'll be in Venice, buy a 24,48, or 72 hour (30E) pass. You can hop on on and not have to cue up for a ticket or buy one on board when the booth isn't open!
Depending on where you are going, lines 1 and 82 go towards San Marco. Ask your hotel which stop is best.

Where exactly in Venice are you staying ? There is an Alilaguna boat from the airport which stops at several places - 2 slightly different routes include stops near San Marco, at the Zattere and sometimes the Lido, plus probably other stops I've forgotten. If you are staying near one of its stops that would save a 2 leg journey of land bus + vaporetto. It costs €12.
The land bus departs from right opposite the terminal. The Alilaguna boat & private water taxis (c.€80-100) depart from docks c.10 minutes walk away, but the route is well signposted and trolleys are available (you need a €1 coin).
