Impossible to answer! If you had asked 3-4 best things to see/do. So I'll give you my top few which I enjoyed.

Barcelona: Gothic quarter, Las Ramblas, Picasso museum.

Rome: All of ancient Rome! Roman forum/colosseum, Campo di Fiori, Piazza Navona, Trastevere, Spanish steps, Pantheon, any outdoor cafe, Vatican.

Naples: National Archeological museum; there's an excavation going on in the heart of Naples (I'd have to get you the name of it) which was absolutely fascinating; eating pizza (the BEST in Italy!).

I haven't been to Cannes and Messina, so hopefully others can help you.

Sounds like your taking a cruise, Fran. If so, take the trip to Taormina from Messina and go see Pompeii while in Naples.

Rome-Sistene Chapel was fabulous (go early or you won't see anything but), wander around and see the fountains and the Colosseum and the Forum if you have time.

Barcelona-Parc Guell(Gaudi's fantasyland creation)and Las Ramblas were my favorites.
