Rabbies small bus tours are often recommended here, I've done one and liked it a lot:

IMO, Rabbies is the best. I've done several tours with them.

looks like most of the longer tours depart from Edinburgh.

In case the OP isn't aware--Edinburgh and Glasgow are only about an hour apart, so I wouldn't limit it to Glasgow departures.

You can also combine 2/3 day tours from Glasgow.

We will be in Glasgow for a conference. But could get ourselves to Edinburgh to start. or start there actually and end in Glasgow..

The Edinburgh tours depart from Rabbies Cafe at Waterloo Place. With the exception of the one day tours, they depart at 9:00. Waterloo Place is practically next door to Waverley Station. There are frequent trains from Glasgow to Waverley but don't cut it too close as some occasionally get cancelled or delayed. It's usually about a 50/55 minute trip.
