It depends if you want to go the quickest route (Autobahn A-8 Salzburg to Munich-By-Pass (exit 95. By-pass A-98(which is not finished all the way) to Autobahn Munich- Lindau (A-96) to Landsberg/Lech (exit 25). South on B-17 to Fuessen. About 300km Depending on traffic,especially around Munich, between 4-5 hrs.
More scenic route:
A-1 Salzburg to exit 115. Bad Reichenhall on B-21 to Lofer; B-178 to Woergl. Autobahn A-12 to Innsbruck/ Zirl (exit 87); B- 177/B-2 to Garmisch-Partenkirchen; B-23/B179 to Fuessen (via Leermoos). About 350 km; because of less autobahn figure on 6-7 hours. All good roads.

I used viaMichelin to map out a scenic route from Salzburg thru Ettal and Wies to Fussen using non-autobahn roads--about 4 hours was the estimate. See if this link will help:
