Any Castle as a castle will have been built before Cromwell and most destroyed by the same. As such most "Castle" hotels are either extended Tower Houses or Victorian built Manors.
As to wheelchair access improvements have been made but Old buildings are not always adaptable. Try sending your request to http://manorhousehotels.com/ see if they can help.

Carton House is not a castle, but a Palladian mansion and certainly wheelchair accessible http://www.cartonhouse.com/
I'd allow 30-40 minutes to get there from Dublin, depending what part of the city you are coming from and assuming not in peak traffic.
Kinnitty Castle's website says it is about 90 minutes away from Dublin http://www.kinnittycastlehotel.com/location.html - that might be too far for you. I'd say it is also wheelchair accessible and is more like a "real" castle.
Can't think of any real castles closer to Dublin unfortunately, and any of the traditional tower houses will presumably not be wheelchair accessible.

Barberstown Castle isn't too far from Dublin. It's really a medieval keep with a more modern building attached - it used to be owned by Eric Clapton! Don't stay there if there's a wedding taking place - the party tends to take over the entire place.
