Help with Duoro Valley

Greetings! I'm planning a trip with my wife to Portugal. I need some help with the Porto - Duoro Valley. We plan 2-3 nights in Porto. We want to go up the Duoro River to the vineyard/winery areas. Is it possible to take the train (or bus) up or back and take a boat the other direction? I see that it is possible, but my searches all come up with packaged tours. Ideally, I want to take the 6 hour boat up to the winery area, stay a night or two in a Quinta and then return via a faster route. Here are my questions:
1.). Are there boats that are comfortable that cruise up the Duoro River and have breakfast/lunch?
2.). Do I need a car in the winery area to see it or get to a good Quinta?
3.). Recommendation for a Quinta? I'm a winemaker (Sonoma County) and would be interested in a working Quinta with a view?
4.). Is 2 nights OK - enough to do, or do I need to rent a car when I arrive via river boat?
