It's been about 3 years, but the car ferry we took was about 7 hours from Pireas to Santorini. We went in April and I got seasick within the first hour and thought I was going to die for the next 6 hours. Rough seas in April I guess. But upon arrival in Santorini, I forgot about my seasickness and it was worth the trip there.

It's a matter of personal preference. I prefer flying, but I've been there many times and for me the approach by ferry is no longer worth the time lost. Aegean Airlines has the best prices, and if you're lucky you might be able to get one of their discounted tickets.
As to calm waters: If you're on a large car ferry you'll hardly notice if the sea's not smooth.
