Hello Helen,
The fastest journey time is 1hour 47mins by Eurostar, with a change of train at FOLIGNO.
This leaves Rome at 07.37 and you arrive at Assisi at 09.24.
The 2nd class fare is 27.700 LIRE
Alternatively you can take an IR (inter-regional) train leaving Rome at 08.14. This is a direct train to Assisi/Perugia, arriving at Assisi at 10.32.
The fare would be about 20.000 LIRE

Hello Diane,
Yes, Assisi is DEFINITELY worth a day-trip from Rome! It's one of my favourite places in Italy, - not to be missed! Make an early start to make it worthwhile, - there's so much to see...
Assisi itself is about a 20-minute bus ride from Assisi railway station. There's a connecting bus service.
N.B. When you get off the train don't hang around, - go straight out to the bus. If you miss it you'll have a long wait! The driver will send you into the station bar to buy your tickets, - but the main thing is to speak to him first so he knows to wait for you.
