Lake Louise is about a 9 hour drive from Vancouver through the mountain ranges. It is a scenic drive however it would take approximately 3 days (2 driving, 1 visiting) out of your 6 days in Vancouver. Lake Louise is much closer to Calgary-just less than a two hour drive. There is plenty to see in the Vancouver area over a 6 day period and I would not recommend taking the time to go to Lake Louise this time. If you wanted to fly into Vancouver, drive across stopping in Lake Louise (and Banff) and flying out of Calgary, you could do it that way.

Vancouver to Lake Louise is about 900 km (565 miles) first through the traffic in the Vancouver area and then across several mountain ranges until you get to the Rockies.
If you make stops along the way (for meals, and refueling the vehicle) it'll take more than 9 hours one way.
What time of the year are you planning to travel? Weather and road conditions could also be factors (that slow you down), as will any road construction along the way.
