What are your interests? There are the beautiful Royal Botanic Gardens as well as Fitzroy Gardens, where you can visit a replica of Captain Cook's cottage. The Melbourne Museum is worth a few hours. You can take a nice boat ride on the Yarra River. There's also plenty of sporting and cultural events going on in the city. For day trips, I enjoyed going out to the Great Ocean Road for the lovely coastal scenery and rock formations as well as a visit to Philip Island to see little penguins. If you enjoy wine, the Yarra Valley isn't far away either. I could easily spend a couple of weeks in Melbourne and not run out of things to do.

Two great art galleries. There's the Victorian National Gallery in St Kilda Road, just across the Yarra river - excellent collection. Also the Ian Potter Centre in Federation Square on the corner of Swanston and Flinders Streets (specialising in Australian art).
Good market at the Queen Victoria market - for atmosphere, stalls and good food. Visit the Block Arcade (off Collins Street) and have tea and cakes at the Hopetoun Tearooms. Lots of good shopping in Melbourne CBD.
Take a coach tour (or hire a car) and drive through the Yarra Valley winery area and visit Healesville Sanctuary to see Australian animals and birds including platypuses.
Take the ferry to Williamstown and have lunch, or stay in town and have lunch at one of the many eating places at Southbank overlooking the river.
