Two nights? Are you not interested in the temples? Two nights is just one full day. If you wanted to just visit the major temples it would take three full days.
You don't need a tour. You can hire a car and driver and guide for your day there.
It's difficult to recommend a hotel with no guidance about what you are looking for. Take a look at www.agoda.com as they have a good variety of hotels in Siem Reap and very good prices.

Actually, it would be 2 days as we intend to leave Bangkok early AM and have 1/2 day, then 1 full day, then another 1/2 day and return to Bangkok in the PM.
Looking for very comfortable but not extravagant hotel-
3 or 4 star.

If you are looking for a low cost option then Earthwalkers http://www.earthwalkers.no/index.php
is a great place to stay. The owners are very involved in local charites as well. My friends stayed last year and said it is still great.
A double with air is $20 and we arranged a car and driver from there as well.

The Bopha Angkor is a nice mid range hotel with a lovely pool and within walking distance of the downtown area. Price range 50-100

We are arriving in Siem Reap mid-November.
Checked out several hotels. We are staying at the Atanue......on the East Bank of the river......but convenient....and the price is under $38/night.

I stay at Shining Angkor hotel About 2 km from town, but, for me, it has comfortable beds, good air conditioning, free wireless internet in the room. Around US $35 a day or so. Will be there in September for my third stay in 12 months
