I'd suggest you use www.kayak.com to search for the best routes from your location to Cape Town. "Best" for you might be routing, length of journey, or price... varies from person to person.

Ditto what ShayTay says. Some prefer to go to Europe, even stop there for a night or two, then continue on down to CT; others prefer to "get it over" as quickly as possible. I don't believe there are any nonstops from the US to CT and I don't believe there are many from Europe. Personally, I like South African Air 204 which flies from JFK in the a.m. to JNB, arriving in the early a.m. (about 8-9 a.m.) at JNB which gives you a whole day to overcome lag or to easily continue on to CT.

From the US you can fly direct only with Delta from Atlanta nonstop to JNB or South African Airways (SAA) from JFK nonstop to JNB. SAA also flies from Washington Dulles but that flight stops in Dakar, Senegal in both directions. From JNB you take a flight or train to CPT. There are nonstop flights to CPT from several European cities. From LHR with BA and Virgin, from AMS with KLM, from CDG with Air France, from FRA with Condor (twice a week, they are part of the Thomas Cook Group, a leisure airline), and from MUC with Lufthansa. There may be others but these are the main ones.
