The Ciudad Perdida is not your run-of-the-mill hike -- it's pretty demanding. You really only take the clothing and gear you will need for a few days, all of which should fit into a normal camping backpack. That being said, I guess if you have enough money, a personal porter can be made to appear but that is something you should ask before you you book.
If you're the gambling type then by all means forgo the anti-malarials. I can't speak for anyone else, but I'd rather take the chance of having bad dreams for a few weeks over contracting an incurable blood disease for the rest of my life.
