Define "port" in this instance. Valetta overlooks Grand Harbour on one side and Sliema & The Ferries on the other; if by "port" you mean the working docks, these are in the area known as "The 3 Cities", on the opposite side of Grand Harbour from Valetta - catch a bus from the city gate if you want to visit.
Alernatively, if you just want to look at boats & ships, find a nice cafe on the city walls & chill.

Are you cruising, by any chance? Just thought that might be the reason for mentioning the port.
I took a horse-drawn carriage ride from the port. I had the driver drop me off in the "downtown" shopping area at the end of the tour, and then took a taxi back to the port. I could have walked back down, but I was carrying packages and thought the ride would be better!

If you mean the port where the cruise ships come in, it can be walked, as we've done it, but it takes a while. Allow about 45 mins and it's mostly uphill. There's nothing particularly interesting to see between the port and town but once you reach Valletta there is heaps to see. There are buses all over Malta, you can pick up a bus map at the tourist info, just inside the Valletta city walls on the right. It lists all the bus numbers and which bus goes where. You can always hop on a horse and buggy which seem to be always around where the tourists are.
If you mean the Grand Harbour, Valletta overlooks this and Valletta is easily walkable, though some parts can be a bit steep and have stairs instead of a roadway.

The port isn't very far away, a nice walk uphill but with some lovely sights along the way.
Just a message for carrom- I'm not sure that you'll be able to rent a motorbike in Malta, but if you do, you must be really careful! The roads are terrible there, and Maltese bikers cannot believe that anyone would actually choose to bike there unless they had no choice!! They all take the chance to go 'overland' -ie. to Europe as often as possible!
