You can book in advance on the web, at the site listed by the previuos poster. However, if your itinerary is 'loose', you can call their phone res line when in Rome. I was a little unsure of whether I'd do it correctly when I phoned from my hotel room but they had a "press 1 for English" etc type of phone menu and it was super easy, English speaking operators - I was there in October, called at 11AM on a Tuesday and was able to get a 1PM visit time. Depending on time of year you're traveling, you might not want to cut it *that* close, but the phone procedure was simple enough. A few other visit tips - they like you there to check in an hour in advance of your entry time, so bring something to read and maybe a little snack to enjoy on the villa grounds. And: if you're someone who loves to see art without a big crowd of people in your immediate vicinity, when you get inside, hightail it up to the second floor and explore there while everyone is doing the first floor...then head down as they make their way up to the 2nd floor.
