Oak Bluffs and Edgartown are the main towns on the island and are extremely walkable. Your ferry from Falmouth will bring you into Oak Bluffs so you could start the day there and then take the public bus to Edgartown or vice versa. Both towns have tons of restaurants, shops and even in-town beaches. In Oak Bluffs, you can see attractions like the Gingerbread Cottages and Flying Horses Carousel. In Edgartown, you can visit memorial wharf, the harbor area The Carnegie museum or just stroll around the gorgeous streets and look at the amazing houses!
If you really want to see everything on your day trip, you might be better off taking a tour of the island. The Aquinnah Cliffs and fishing village of Menemsha are two very popular spots but are on the other side of the island. Public transport does go there but you would spend your whole day getting there and back. There are public tours departing from Oak Bluffs that take 3 hours and cost around $60 per person. Private tours are more flexible and can bring you wherever you like but they'll start around $400 for 3-4 hours.
