Is it possible & what's best way to fit in Monticello in one day (no night) during my WashDC stay Oct 20-24?

OK, no car.
By Metro and bus: Greyline, Tour Mobile or public transport (metro to Huntington Station on Yellow line, Fairfax Connector bus directly to Mt Vernon)

You mentioned a boat cruise to MV above. One of the companies (Spirit Cruises) included on MV's website books trips through the end of October, weekends only.
Regardless of transportation choice, it will be almost impossible to get to/from and see MV in a morning. Might work if you had a car, got there at opening time and did a cursory look around. There's a fair amount to see there. Realistically, I'd give the door to door trip 5 to 8 hours.
You could do the monuments at night. Tourmobile and trolley tours do evening tours of the monuments. It'd be a long day. There's so much to see!
