Well, I've been here 47 years and I'm still touring... :)
It really depends on what you like, of course. The historic area alone is easily two days, and then there are the various museums around the city, accessible by walking/public transportation. There are some great walking tours, including a chocolate tasting one and some by the Foundation for Architecture, plus the possibility of exploring the outer neighborhoods, like Chestnut Hill or Manayunk. Day trips out of the city that are popular include Longwood Gardens, the mansions of Delaware, Valley Forge, or even Lancaster (the Pennsylvania Dutch culture).
It's a wonderful city for walking, and we could easily give you a week's worth of activities/itineraries. www.visitphilly.com is a good place to start looking, just to get a feel for the area.
