JFK from manhattan on marathon sunday
I'm traveling from JFK at 6pm on marathon Sunday. Need to get taxi from upper east side. Will all roads be closed?

If you google "NY Marathon road closures" you'll find that all roads that were closed for the marathon will be open by 4:30PM.

The Triboro Bridge, which is the one you cross from the UES to get to JFK, does not close for the marathon, and it can be reached completely by city street routes that will not be closed that day. By 6pm almost everything to do with the marathon will be over, so I doubt you will have any delays. The only time you might have difficulties is if you are staying on Fifth Avenue.

After writing the above I began to think because what I wrote isn't strictly accurate. First Avenue is actually the largest part of the Manhattan route, and you have to cross 1st Ave to get to the Triboro Bridge. By 6 the race is basically over, but I suppose there could still be people running up 1st Avenue, and that could cause traffic problems for people going onto the bridge.
