According to www.trenitalia.com, trains will run on May 1 between Rome and Venice.

But don't count on local public transportation in Rome (I have read in a guidebook that the buses do not run on May 1; I know from experience that they did not in Siracusa last year). I have no idea, unfortunately, whether the vaporetti in Venice run on May 1.

You can try to find the information on the two following Web sites:

www.actv.it (Venice)
www.atac.roma.it (Rome)

May 1 is a big holiday in Italy. When I was there last year in the Cinque Terre (considerably smaller than a Rome or a Venice!) the trains and busses were not running into/out of the CT nor in neighboring La Spezia. To play it safe, you might have to bump your travel date up or down by 1 day if the transportation is lacking or nil that day.
