The way to figure this out is by going to the Deutsch Bahn website - http://www.bahn.de/i/view/USA/en/index.shtml .

As I recall, you can get to Fussen, but you have to change trains on the way, which is easy to do. From Fussen you can get a bus to Neuschwanstein. Salzburg is a direct trip from Munich, which takes about 2 to 2.5 hours, I've done it several times. I think the only way to get from Fussen to Salzburg by train is to go back through Munich.

As far as going by bus, there are probably companies that offer that trip, but I'm not really familiar with bus travel in Germany.

Neuschwanstein is not on the route to Salzburg. Rather in opposite direction.

As already mentioned there is another one of King Ludwig II's castles en route: Herrenchiemsee. From the interior anyway the much more interesting one. And also a nice location.

Munich to Prien train station. There are luggage lockers.

From there either walk or take the cute Chiemsee railway (kind of steam tram) to Stock, the harbour of Prien.
Boat from there to Herreninsel island where Herrenchiemsee New Palace is located.

Later continue from Prien by train to Salzburg.
If you use regional trains for Munich - Prien - Salzburg you can do this for cheap on a Bayern Ticket (EUR 26 for 2 persons - assuming your daughter is older than 14 years).

There is a way you can go by train/bus from Fuessen to Salzburg, but it is a bit longer than going via Munich. But it is much more scenic. For schedules, times, etc. on all of the following itinerary, check www.bahn.de. There is an English language version. "Follow the flag", in the right hand top corner of screen

From Munich to Fuessen (Neuschwanstein) some trains are through trains, on some you have to change trains in Buchloe. You can do this trip using the Bayern Ticket discounted day ticket. Limits: On weekdays, trip can't commence before 9AM. You re limited to Regio trains (no IC, ICE, EC), but since that is all that is running between Buchloe and Fuessen, it's no problem. Tiket price: Single person EUR22, each additional person another EUR4, up to 5 people on single ticket.

From Fuessen to Salzburg: Bus to Reutte (short ride), Reutte - Garmisch< train (quite scenic); Garmisch- Innsbruck < train (quite scenic, sit on right side of coach; Innsbruck - Salzburg
You can buy Bayern ticket at kiosk or ticket window at Munich station. The rest of the ticket, at ticket window in Munich station.

I don't remember for sure from my last trip, but I think the answer to your last question is no. You just buy them in the train station the day of travel. There is a ticket office in the train station, and you can either buy them at the ticket windows, or they have a bunch of ticket machines in the ticket office with a staffer there to help you. The machines are usually quicker.
