Train trip to Berlin with a hiking hut expedition along the way?

My husband and I are traveling from late August to early September (2013)--8 nights--and we'd like to go from Amsterdam to Berlin by train. We'll stay in Amsterdam 1-2 nights and Berlin at least 2 nights. Along the train route we'd like to stop somewhere, hike to a German hiking hut (we've heard so much about these) and then hike the next day back to a train that can take us to Berlin. Is this even possible?? We'd really like to hike to a hut that has no car road access. This seems a little daunting to try and figure out and..we've never visited Germany before.
