I plugged the two points into Google Maps. You can do the same. It shows solutions by bus, metro, RER, or a combination. Looks like it would take about half hour and at least one change.

I'm OK changing metros with luggage, but some people aren't. (I'd avoid changing between metro and RER.) In that case, take a taxi.

If you are only in paris one night, I'd probably just stay
near gare we Lyon otherwise I'd stay in montparnasse

You have two options:

1. walk to the taxi stand at the corner of Raspail and blvd de Montparnasse and take a taxi. You can call a taxi if you like but the 5 minute walk to the taxi stand will save you the 4€ dispatch fee.

2. walk to the Edgar Quintet métro entrance for line 6, take it to Bercy and transition to M14 for Gare de Lyon.

As you mention fastest, I would not even consider the bus. At any give time, either option #1 or #2 above could be faster. It depends upon traffic conditions. Either way I would allow a minimum of 30 minutes.

IBIS Hotel opposite Gare de Lyon is a good bet (www.accorhotels.com) - if want to stay near the station.
