The most straight forward trip is to take a Trenitalia train referred by the above link.
Departing Milano Centrale, Arriving Varenna-Esino. The trip should be around 1hr3min.
Varenna is an unmanned station. If you plan to take a train from Varenna, use a ticket vending machine or buy tickets for the future trip also before you hop onto the train from Milano Centrale.

oops, I was looking at another posting. Sorry giving the wrong destination. Enter Milano Centrale departure and Vernazza as destination in the link provided by ellenem. You have to change the train at Genova Piazza Principe or Sestri Levante.

Poster is going to Vernazza in CT not Varenna (I believe). It is an easy train ride from Milan to La Spezia and then change trains to a local that stops at all 5 towns of CT. We took the trip last spring and it was very easy.

To find out how long the journey is, look at one of the railways' websites. www.bahn.de gives schedules for all of Europe.
