As you probably know, they're neighboring sister hotels, members of LHW, to the west of Marbella towards the yacht harbour of Puerto Banús on the Golden Mile and they share some of their facilities.
The Puente Romano is the" newer" of the 2, opened in '76, the Marbella Club having been around since the 50s, built as Prince A. Von Hohenlohe's private estate to house his "jet set" friends. Back in the 70s and 80s it certainly got its share of Hollywood guests who used it as their private club. The M.C. is smaller, with more private bungalows, in other words, more "clubish", and it's also slightly (and very slightly) more expensive. (In low season we can actually afford a night or two at P.R.) I have a Fodor friend who spent her honeymoon at P.R. and loved it, but I know others who wouldn't stay anywhere but the M.C., primarily for its exclusive "pedigree" and its old glamour factor. I think the P.R. attracts a younger crowd.
