Menerbes is a beautiful town, and you could very easily access the other Luberon villages and the countryside. This base also puts you in a central location to reach out up to hour in various directions, which takes you to Vaison and Aix you've identified and also St. Remy, Arles, Avignon.
Most rental houses are rented for a full week, Saturday to Saturday, unless you can strike a deal in the off season. Sometimes people do pay for a full week and not use the entire time, which might be cheaper than a B&B.
Think about how you feel about driving two hours or so round trip to do several days of day trips during a 5-6 day stay. (The scenery is always interesting!) On our first trip to the Luberon (we rented a house in Saignon), we did three day trips and stayed in the Luberon three days... alternating our schedule. It was the best trip we'd ever had. Staying in a rental really does offer a different kind of experience.
