Madrid- what to see, Barcelona?

Madrid- what to see, Barcelona?
Hoping for some help--
Two active adults traveling to Madrid on business in late September. Our time is somewhat limited, we'll have one full day/night to explore and then several hours here and there through out the week. I've booked a segway tour for one morning. Am wondering what to do with the one full day/night. We've talked about seeing Barcelona, but realize that's a long way, makes for an exhausting day. But, feel like we're so close, not sure when we'll get back. The other appealing idea is a bike tour of Segovia. This is a full day trip. Any other ideas? We believe that we'll see museums and sights within the city during the week when we have blocks of time, so would like to get out for that one day.
Also, we're celebrating my 40th birthday and have that evening (which is a Sunday) free to celebrate. An suggestions? Had thought about a Flamenco show, and a nice restaurant. I need to get reservations now. The shows start late- first one at Corral de la Moreria is 10pm. We could do dinner first, not eat there.

Thanks all, we are working on a day trip to Toledo for 9/28. And, for my birthday night we're taking a tapas/wine tour. I'm still hoping to see Segovia and looking for a good tour recommendation. The bike tour didn't work out, they aren't having them to that town next week.
