Looking for advice, please.

My sister and I will be traveling to Europe in October... and we are hoping someone can give us the best advice about the smartest ways to travel, stay, tour, etc. I will be 66 by then, and she is 59. We will stay in Rome for a week (and the hotel is already paid.) The second week is not figured out yet. I am hoping that we can see Neuschwanstein Castle, and visit Salsburg and Munich, and then we will fly out of Zurich to return home.
Is it best to purchase reserved seats on the train? From what I've seen in articles, we would have to get a pass for 4 countries - and for 5 days. Or would it be better to purchase individual tickets?
How much can we really do in a week? And what would you recommend above all?
I thank you from the bottom of my heart for any assistance you can give us!
