And itinerary I tried to tailor to OP's wishes could well work - a little huried for some folks but let OP be judge of that.

I'm not sure if your family has flown overseas but we don't all adjust to jetlag the same. So your kids may not be ready to move to a new site after 2 days. Also, it's pretty hard to see the Vatican and the Colosseum in one day,so are you willing to pass on the Colosseum? Or, one possibility, from reading your schedule, it appears you are arriving in Rome on Friday. I think that is the day the Vatican Museums have night hours of 7-11 pm. So you might be able to see the Vatican Friday night, and St. Peter's Saturday morning, then the old parts of Rome and the Colosseum Saturday afternoon.
Take a train between Naples and Florence, and between Florence and Rome, and that becomes part of the experience. In both Florence and Rome you will want to maximize your time by where you stay. Are you wanting to pre-book tickets to any of the sites? It might be a good idea.
If you want to stay near the Vatican, that might help. Take at taxi from Termini train station to your hotel, eat and walk over. I have stayed at the Hotel Alimandi, and have had other relatives and friends stay there on subsequent trips. Small rooms, breakfast, airport shuttle. Another option I nearly booked and read good things about is Hotel Sant Anna. Just a few blocks from the Vatican. Check Trip Advisor or similar for recent reviews.
