I agree with most posters - you are doing way too much driving. Day trip #2 is the most reasonable, don't remove the Pienza - San Quirico d'Orcia trip, it's possibly the most beautiful area in Italy. I would also do Sienna as my 'big city' day trip excursion. On your last day, you might want to devote more than 2 hours to Orvieto, a very interesting large hill town.
Since this is your first trip to Italy, just a FYI about driving in Tuscany: Driving is not too difficult if you go in with the right mind set. You will often have tailgaters on the narrow, shoulderless roads, and you most likely will get lost a few times. Also, driving into a city like Sienna can be very frustrating and confusing, most streets are narrow, one way and winding, and can change names from block to block. Have the most detailed maps you can find, and a good navigator!
