And there aren't any "must-do's." There are only things YOU want to do.

Two more reasons to hit up the guidebooks, maps, and internet.
Time DOES permit a day trip to Venice. There are 16 days in Spain with no itinerary. So why would you say that time does not permit a day trip to Venice?

A day trip to Venice from a base in Spain makes absolutely no sense at all, particularly with your budget. But time does permit a day trip to Venice!

Do you have tickets already? Why not fly into Venice for three days, then to Barcelona, and home from Madrid. Buy multi-city tickets, into one city, home from another.

To go to Venice from Barcelona, you might not be using main airports, so would use more travel time getting to and from regional airports. RT in one day would eat up at least 8 to 9 hours, and not be a pleasant first time in Venice. If your travel partner is thinking of Romantic Venice, it won’t be in a horribly rushed day. Much of Spain is also very romantic.

Get a Couple of up to date Guide books for just Spain and Rick Steve’s Europe through the back Door. Fodor’s is great for general info. National Geographic has good info on various sites, plus pictures. Pick up a paper map of Spain and one of Europe, so you can open them up and get the big picture. It is old school, but really fun to spread it out on a table and get a sense of locations and sizes of places.
Spain is a big country. You will see only a bit in 16 days. Two nights in a place gives you one day, three nights gives two days, etc. Do not count travel days as days sightseeing. You can go at a fast pace for a week or so, but will likely need a down day now and then.

There are many, many other good plans, different places to add, Malaga, caves, a beach in the South, Jerez, white villages, Cadiz, Salamanca, etc. It depends on your interests! These are minimum times. All could be extended for a day trip, another night, hiking, concerts, etc.
Basic plan
Arrive Barcelona, 4 nts, 3 full days (that is really short. . . ., would want more for day trips.)
Fly to Granada, 2 nts, 1 full day, need more for hiking
Travel to Seville, 4 nts, 3 full days
Train one hour to Córdoba, 1 nt, most of day
Travel to Madrid, 3 nts, 2 &1/2 days, use one day as a day trip to Segovia?
Train (1/2 hour) to Toledo, 2 nts, 1 day
Travel to airport

Already booked our ticket. Multi city or one way tickets were a lot more than our r/t to Barcelona. We're going to spend the first 7 days in Barcelona. The other 9 days are still up in the air.
