Best route of travel to the Cinque Terra from Palermo

Spending a week in Sicily with family then on to Cinque Terra. What is the best route to do so?

Fly from Catania or Palermo to Pisa or Genoa, train to Cinque Terre.

See skyscanner.com. Non-stop to Pisa from Palermo. Get to Pisa Centrale, buy train tickets to the village of your choice from a Trenitalia machine, validate before boarding the train using one of the little stamping machines.

Just as an aside, have you considered a coastal destination in Italy that isn't so far away? For example, the Amalfi coast offers hiking which is even better and is easier to reach from Sicily. Just a thought.

Take the ferry (comfortable cabins, restaurant, sun decks, etc.):
Palermo dp 23.59 - Genoa ar 20.00,
Palermo dp 23.30 - Livorno ar 17.30, connecting trains
Monterosso dp 18.46, 19.31, 20.47 - Monterosso ar 21.27, 22.43, 23.34

It is no more or less difficult to reach the Italian Riviera from Palermo than it is to go to the Amalfi coast from Palermo. As for hiking, it depends on what you want to do and see personally. It's subjective.
